Adam Savage and Jamie Hyneman are the original MythBusters, and initially explored all the myths of the series using their common experience with special effects. The two work at Hyneman's effects workshop, M5 Industries; they make use of his staff, which often works off-screen, with Hyneman and Savage usually shown doing most of the work at the shop. One of the series' gimmicks is the interaction between Savage and Hyneman, which is similar to a double act, wherein Hyneman plays the straight man and Savage is the comic relief. The show is narrated by Robert Lee, though in some regions his voice is replaced by a local narrator.

On the show Kari Byron, the Eye Candy, is featured along with fellow Build Team members Tory Belleci and Grant Imahara and also assists as co-hosts for Adam Savage and Jamie Hyneman in testing the plausibility of various myths. She became involved in the show after persistently showing up at Hyneman's M5 Industries workshop in a desire to get hired at his company. On her first day as a paid employee, she was asked by Jamie and MythBusters producer Peter Rees to help out with the "vacuum plane toilet" myth, and she was asked to model her buttocks for a cast: a job which would later have her comment (jokingly) that she was hired "for her butt". She was offered $100 for the task and accepted because she did not think that anyone would see it; "Now you Google my name and it's the first thing that comes up.", she said. She was given a more prominent role in the show starting with its second season. Not having had a long history in show business, at first she found it hard to act naturally with this more visible position but gradually became more used to it.

Tory Belleci began work on Mythbusters in 2003 doing work behind the scenes and in the second season was featured as part of the show's build team and got on screen credit in the shows third season. He is often considered by fellow MythBusters Kari Byron and Grant Imahara to be more of a daredevil and as such often performs the more dangerous stunts when testing a myth.

Grant Imahara joined MythBusters on the invitation of friend and occasional employer Jamie Hyneman and former ILM colleague Linda Wolkovitch (now an Associate Producer on MythBusters). He joined as the third member of the Build Team, replacing former MythBusters welder Scottie Chapman. His colleagues often jokingly refer to him as the geek of the Build Team, and he often makes the robots that are needed on the show and otherwise specializes in operating the computers and electronics for the myths.
